Overview

TRMT61B is a mitochondrial RNA methyltransferase enzyme encoded in the nuclear genome but localized primarily in mitochondrial matrix, where it catalyzes the formation of N(1)-methyladenine (m^1A) at position 58 of specific mitochondrial tRNAs and at position 947 of mitochondrial 16S rRNA[1][3]. These methylation events are critical for proper mitoribosome structure, mitochondrial translation, and protein synthesis. Loss of TRMT61B impairs mitochondrial respiration and can decrease tumor cell proliferation, especially in highly aneuploid cancers, linking it to mitochondrial genome regulation and cellular metabolic homeostasis[5]. Elevated expression is associated with tumor aneuploidy, and abnormal TRMT61B levels may play a role in cancer and neurodegenerative disease[1][3][5].
