Overview

Trophozoite exported protein 1 (TEX1) is a protein expressed by the Plasmodium falciparum parasite during the erythrocytic stage of malaria infection (UniProt: Q8I6R7). It is localized to the Maurer's clefts, which are essential organelles for the trafficking of parasite proteins to the host erythrocyte membrane (PubMed: 25406331). The P27A peptide is a 27-amino acid sequence derived from a highly conserved, unstructured region of TEX1, identified as a target for antibodies that inhibit parasite growth (PubMed: 21423664). As a therapeutic target, P27A is utilized in vaccine development to elicit protective humoral and cellular immune responses against the blood-stage of the parasite. Clinical trials have demonstrated that P27A-based vaccines are safe and immunogenic, inducing antibodies capable of inhibiting parasite multiplication in vitro (PubMed: 28973454). The target is significant due to its high conservation across various P. falciparum strains, which may overcome the challenge of antigenic diversity in malaria vaccine design (ClinicalTrials.gov: NCT01945112).
