Overview

Trophozoite-expressed protein 1 (Tex1) is a protein expressed by the malaria parasite Plasmodium falciparum during the asexual blood stage of its life cycle (UniProt Q8I6R7). It is localized to the surface of infected erythrocytes, where it is involved in the remodeling of the host cell membrane (Olugbile et al., 2011, Vaccine). The P27A vaccine is a synthetic peptide candidate based on a highly conserved, intrinsically disordered region of the Tex1 protein (Nebie et al., 2015, PLOS ONE). Vaccination with P27A aims to induce high titers of specific antibodies that can recognize the native Tex1 protein on the surface of infected red blood cells. These antibodies are intended to mediate parasite clearance, possibly through antibody-dependent cellular inhibition (ADCI) or by interfering with the parasite's ability to sequester in host tissues (Audran et al., 2015, Human Vaccines & Immunotherapeutics). Clinical evaluation has shown that the P27A vaccine is well-tolerated and capable of inducing robust immune responses in humans (Nebie et al., 2015).
