Overview

Tryptophan 2,3-dioxygenase (TDO; EC 1.13.11.11) is a heme-containing enzyme that catalyzes the first and rate-limiting step in the kynurenine pathway of tryptophan metabolism, converting L-tryptophan to N-formyl-L-kynurenine. This reaction is crucial for regulating systemic tryptophan levels and has significant implications for immune and neurological homeostasis. TDO activity is tightly regulated by tryptophan levels: high tryptophan stabilizes tetrameric active conformation, while low tryptophan destabilizes tetramers leading to degradation. Dysregulation or aberrant expression has been implicated in digestive system diseases and cancer, making it a promising therapeutic target.
