Overview

Tsix transcript, Xist antisense RNA (TSIX) is a long non-coding RNA that is transcribed antisense to the Xist gene at the X-inactivation center (XIC) on the X chromosome[2][3]. Its primary role is as a cis-acting negative regulator of Xist expression, thereby controlling the process of X-chromosome inactivation in female mammals. TSIX is expressed from the active X chromosome and prevents Xist upregulation, thereby blocking the initiation of inactivation on that chromosome[3][4]. This antisense RNA is non-protein-coding and functions through mechanisms that may involve chromatin modification and DNA methylation at the Xist promoter[2]. The dynamic and mutually exclusive expression of Tsix and Xist is essential for dosage compensation in female mammals. There is no evidence to date that TSIX is a direct therapeutic target, biomarker, or safety concern.
