TTK protein kinase, also known as Monopolar Spindle 1 (MPS1), is a dual-specificity kinase that plays a critical role in the spindle assembly checkpoint, ensuring proper chromosome segregation during mitosis (Source: UniProt P33981). TTK is significantly overexpressed in a wide range of human malignancies, including lung, esophageal, and breast cancers, while its expression in normal adult tissues is restricted primarily to the testis (Source: PubMed 18483465). The TTK antigen epitope presented by HLA-A*2402 is a specific peptide fragment of the TTK protein that is processed and displayed on the cell surface by the HLA-A*2402 MHC class I molecule. This peptide-MHC complex acts as a target for immunotherapy, allowing the immune system to distinguish malignant cells from healthy ones. Therapeutic approaches, such as the OTS111 peptide vaccine, aim to elicit a cytotoxic T lymphocyte (CTL) response specifically against cells presenting this epitope, thereby inducing tumor regression in patients who are HLA-A*2402 positive (Source: PubMed 25193415). By targeting this specific complex, drugs can induce tumor-specific immunity while minimizing damage to normal tissues that do not express the TTK protein (Source: PubMed 24633745).
