Overview

The colchicine binding site is a distinct pocket located at the intradimer interface between alpha- and beta-tubulin, primarily involving the beta-tubulin subunit (UniProt: P07437). Tubulin is a globular protein that polymerizes into microtubules, which serve as structural components of the cytoskeleton and are vital for cell division, intracellular transport, and motility (PubMed: 22435312). Binding of ligands to this site prevents the tubulin heterodimer from transitioning into the straight conformation required for microtubule assembly, thereby acting as a microtubule-destabilizing agent (PubChem: CID 6167). This disruption leads to mitotic arrest and subsequent apoptosis, making it a significant target for anti-cancer drug development (PubMed: 24511911). Beyond oncology, the site is the primary target for colchicine, a drug used to treat inflammatory conditions like gout and pericarditis by inhibiting neutrophil activity and inflammatory signaling (StatPearls: NBK431102). Therapeutic challenges include a narrow therapeutic index and potential for multi-drug resistance mediated by beta-tubulin isotype expression or efflux pumps (PubMed: 33806311).
