Tubulin beta chain (TUBB) is a fundamental structural protein that polymerizes with alpha-tubulin to form microtubules, which are essential for maintaining cell architecture, facilitating intracellular transport, and enabling chromosome segregation during mitosis (UniProt P07437, PubMed: 22403158). The specific target site located at the α/β-tubulin heterodimer interface is known as the colchicine binding site, a deep pocket where ligands interact with both subunits to prevent the curved-to-straight conformational transition required for microtubule assembly (PubMed: 24529408). Drugs targeting this interface act as microtubule-destabilizing agents, leading to cell cycle arrest in the G2/M phase and subsequent apoptosis (StatPearls: NBK431102). This target is clinically significant for treating inflammatory conditions like gout and is a major focus in oncology for the development of antimitotic and vascular disrupting agents (PubMed: 28810910). However, therapeutic use is often limited by toxicities such as peripheral neuropathy and neutropenia, as well as the development of resistance through the expression of specific tubulin isotypes like TUBB3 (PubMed: 15138460).
