This target entry represents a combination of three prominent tumor-associated antigens (TAAs) frequently targeted in cancer immunotherapy: NY-ESO-1 (New York esophageal squamous cell carcinoma 1), MUC1 (Mucin 1), and MAGE-A3 (Melanoma-associated antigen 3). NY-ESO-1 and MAGE-A3 are members of the cancer-testis antigen (CTA) family, which are typically expressed only in germ cells but become aberrantly re-expressed in various malignancies, making them highly specific targets for T-cell based therapies (PMID: 29070616, PMID: 27114454). MUC1 is a transmembrane glycoprotein that is overexpressed and aberrantly glycosylated in many epithelial cancers, where it promotes oncogenic signaling and contributes to immune evasion (PMID: 25113845). These antigens are often utilized together in multi-valent vaccines or adoptive cell transfer strategies, such as TCR-T or CAR-T therapies, to address tumor heterogeneity and reduce the likelihood of immune escape. While these targets offer significant therapeutic potential, challenges include the risk of cross-reactivity with healthy tissues—most notably seen in MAGE-A3 trials involving cardiac Titin—and the immunosuppressive environment of the tumor (PMID: 23733940, PMID: 31515355).
