The tumor-associated Mucin 1 (MUC1) glycopeptide is a distinct neoantigen characterized by the presence of truncated O-glycans, such as the Tn (GalNAc) or Sialyl-Tn (STn) antigens, attached to the MUC1 protein backbone (Nath & Mukherjee, 2014). In healthy epithelial cells, MUC1 is a heavily glycosylated transmembrane protein that protects surfaces with long, branched carbohydrate chains; however, in carcinoma cells, the glycosylation process is disrupted, often due to the downregulation of the COSMC chaperone, leading to the exposure of the underlying peptide core and short sugar chains (Beatson et al., 2016). This specific glycopeptide configuration is highly tumor-specific and is found across a wide range of adenocarcinomas, including breast, ovarian, and pancreatic malignancies. Because these truncated glycoforms are absent or inaccessible in normal tissues, they serve as high-affinity targets for therapeutic intervention. Current drug development strategies targeting this antigen include monoclonal antibodies like Gatipotuzumab, which recognizes a carbohydrate-induced conformational epitope, as well as glycopeptide-based vaccines and CAR-T cell therapies designed to induce a potent immune response against malignant cells (Glycotope, 2023).
