Tumor cells in situ refer to malignant cells located at their site of origin within the body. In local cytotoxic chemotherapy exposure, chemotherapeutic agents are administered directly to the tumor site to maximize their tumoricidal effect while limiting systemic toxicity. This approach exploits the cytotoxic nature of classic chemotherapy agents that kill rapidly dividing cancer cells, but since these drugs are not selective for molecular targets, they can also affect normal proliferating cells unless their action is restricted through local delivery[1][4][9]. Unlike targeted therapies, which interact with specific receptors or pathways, classic cytotoxic chemotherapy broadly inhibits cell division and survival by damaging DNA/RNA or blocking mitosis.
