Overview

Tumor necrosis factor (TNF) is a pleiotropic pro-inflammatory cytokine secreted mainly by macrophages in response to infection, injury, or immune activation[2][5][8]. TNF-α is the most well-studied form, exerting its effects via two main receptors (TNFR1, TNFR2), and plays a central role in mediating inflammation, cell death, survival, and immune system regulation[2][3][4][6]. Many autoimmune and inflammatory diseases are associated with abnormal TNF activity, making TNF (the protein, not its mRNA) a crucial therapeutic target for several biologic drugs[5]. The mRNA for tumor necrosis factor is involved in regulated translation and stability, but is not a classical drug target except in some gene expression-inhibiting approaches (antisense, RNAi)[2][7].
