Overview

The Tumor necrosis factor receptor superfamily member 11b (TNFRSF11B) mRNA 3'-untranslated region (3'-UTR) is a critical regulatory segment of the messenger RNA that encodes osteoprotegerin (OPG) [PMID: 9144294]. OPG serves as a secreted decoy receptor for the Receptor Activator of Nuclear Factor Kappa-B Ligand (RANKL), effectively preventing RANKL from binding to its receptor RANK on osteoclast precursors, which inhibits bone resorption [PMID: 10359812]. The 3'-UTR contains specific sequences that serve as binding sites for microRNAs (miRNAs) and RNA-binding proteins, which post-transcriptionally control OPG expression by modulating mRNA stability and translation [PMID: 25650543]. Dysregulation of this regulatory region is linked to various pathologies, including osteoporosis, Paget's disease of bone, and vascular calcification, where the balance between bone formation and resorption is disrupted [PMID: 15355921]. While no drugs targeting this specific mRNA region are currently approved for clinical use, it is an active area of research for RNA-based therapeutics, such as antisense oligonucleotides and miRNA mimics, aimed at restoring OPG levels to treat metabolic bone disorders [PMID: 30639594].
