Overview

Tumor protein p53 (TP53) is a master regulatory transcription factor and tumor suppressor often called the "guardian of the genome" due to its essential role in preventing genomic instability (UniProt: P04637). It is activated by various cellular stresses, including DNA damage, hypoxia, and oncogenic signaling, subsequently inducing genes that mediate cell cycle arrest, DNA repair, senescence, or apoptosis. Mutations in the TP53 gene are the most frequent genetic alterations in human cancer, leading to the loss of these critical tumor-suppressive functions and contributing to therapeutic resistance (PubMed: 15549032). SCH-58500 is a recombinant, replication-deficient adenoviral vector (Ad5RSV-p53) designed to deliver the wild-type human p53 gene into malignant cells (PubMed: 11033161). By restoring functional p53 expression, SCH-58500 aims to re-establish the p53 signaling pathway, thereby promoting programmed cell death and increasing the sensitivity of tumors to chemotherapy and radiation (NIH: National Cancer Institute).
