Overview

Tumor protein p63 (TP63) mRNA is the messenger RNA transcript of the TP63 gene, which encodes a critical transcription factor belonging to the p53 family. It serves as a master regulator of epithelial development, limb morphogenesis, and the maintenance of adult stem cell populations in stratified squamous tissues. Through alternative promoter usage and splicing, the mRNA produces two major classes of isoforms: TAp63, which contains a transactivation domain and typically acts as a tumor suppressor, and DeltaNp63, which lacks this domain and often functions as an oncogene. In various squamous cell carcinomas, DeltaNp63 is significantly overexpressed and acts as a lineage-survival factor, making its mRNA a high-priority target for gene-silencing therapies. Experimental approaches such as small interfering RNAs (siRNAs) and antisense oligonucleotides (ASOs) are being explored to selectively degrade TP63 mRNA and inhibit tumor growth. However, the therapeutic window is narrow because p63 is essential for normal skin integrity and wound healing, and its systemic loss can lead to severe ectodermal dysplasias. Additionally, the complexity of targeting specific isoforms while sparing others remains a significant technical challenge in the development of RNA-based therapeutics for this target.
