Overview

Tumor suppressor candidate 7 (TUSC7) is a long non-coding RNA (lncRNA) of approximately 2.1 kb encoded at chromosome 3q13.31. It acts as a tumor suppressor in various cancers by regulating cell proliferation, apoptosis, migration, and invasion. TUSC7 functions, in part, by sponging oncogenic microRNAs, including miR-23b and miR-211, reciprocally inhibiting their activity. Its expression is typically downregulated in several malignancies, and reduced TUSC7 levels are associated with increased tumor aggressiveness and poor patient prognosis. TUSC7 is also a direct transcriptional target of the tumor suppressor p53. No approved drugs directly target TUSC7, but preclinical studies suggest its modulation may be a valuable approach for tumor suppression and as a prognostic biomarker.

Mechanism of action

TUSC7 acts as a molecular sponge for oncogenic microRNAs including miR-23b, miR-211, and miR-10a, leading to decreased proliferation, migration, and increased apoptosis in cancer cells. It is also involved in p53 pathway regulation, as TUSC7 is regulated by p53 and mediates some p53-dependent tumor suppressor activity in certain contexts.
