Overview

Tyrosinase-derived melanoma antigen epitopes are specific peptide fragments originating from the tyrosinase enzyme, a key protein in the melanin synthesis pathway (PMID: 8389191). These epitopes are processed intracellularly and presented on the surface of melanoma cells by Major Histocompatibility Complex (MHC) molecules, specifically Human Leukocyte Antigen (HLA) class I and II (PMID: 7524369). As tyrosinase is a lineage-specific differentiation antigen highly expressed in melanoma, these peptides are utilized as targets for various immunotherapeutic approaches, including cancer vaccines and adoptive T-cell transfer (PMID: 21148347). By training or engineering the immune system to recognize these epitopes, clinicians aim to induce a targeted cytotoxic T-cell response against tumor cells (PMID: 15150569). However, because tyrosinase is also expressed in normal melanocytes, therapeutic targeting can lead to autoimmune-like side effects such as vitiligo or ocular inflammation (PMID: 11748450). Monitoring HLA status and tyrosinase expression levels is critical for patient selection in clinical trials targeting these epitopes (PMID: 16467100).
