Overview

Tyrosine-protein kinase Fyn is a 59 kDa non-receptor tyrosine kinase belonging to the Src family (SFK), encoded by the FYN gene (UniProt: P06241). It plays a critical role in diverse biological processes, including T-cell receptor signaling, oligodendrocyte differentiation, and synaptic plasticity (PubMed: 21734675). The SH2 domain of Fyn is a conserved protein module that recognizes and binds to phosphorylated tyrosine residues, facilitating the assembly of signaling complexes and the localization of the kinase to specific cellular compartments (PubMed: 12496361). In the context of pathology, Fyn is a key mediator of Alzheimer's disease progression, where it links amyloid-beta oligomers to Tau phosphorylation and NMDA receptor dysfunction (PubMed: 25130974). Additionally, Fyn is overexpressed in several cancers, such as melanoma and glioblastoma, where it promotes epithelial-mesenchymal transition and metastasis (PubMed: 21734675). Therapeutic targeting of Fyn primarily involves small-molecule inhibitors like saracatinib, which has been investigated for its potential to mitigate neurodegeneration and oncogenic signaling (NIH: NCT01864655). Disruption of the SH2 domain specifically is an area of active research to improve selectivity over other Src family members.
