NKG2D ligands are a family of stress-inducible molecules (including MICA, MICB, and ULBPs) overexpressed in various cancers and other pathological states. These ligands bind to the activating NKG2D receptor on natural killer (NK) cells, triggering NK-mediated lysis of the transformed or infected cells. Their expression facilitates immune recognition but is subject to tumor-driven evasion by ligand shedding, downregulation, or immune checkpoint inhibition. Therapeutic exploitation includes antibody-based strategies, CAR cell engineering, and immune checkpoint blockade, although clinical translation faces challenges in selectivity and tolerance.
