The Up-regulated in lung cancer 10-derived HLA-A*2402 peptide epitope (URLC10-177) is a specific amino acid sequence (RYCNLEGPPI) derived from the URLC10 protein, also known as Lymphocyte Antigen 6 Family Member K (LY6K) [UniProt: Q9BYG4]. URLC10 is a cancer-testis antigen that is highly expressed in various malignancies, including non-small cell lung cancer (NSCLC) and esophageal cancer, but is absent in normal tissues except for the testis [PubMed: 21406134]. This peptide is specifically designed to bind to the HLA-A*2402 molecule, a common MHC class I allele in East Asian populations, for presentation to the immune system [PubMed: 22473314]. Upon administration, typically as a vaccine, it induces the activation and proliferation of URLC10-specific cytotoxic T lymphocytes (CTLs) that recognize and lyse tumor cells expressing the antigen [PubMed: 25113111]. Clinical trials have evaluated this epitope both as a monotherapy and as part of multi-peptide vaccine cocktails like S-288310 to enhance anti-tumor immunity in patients with advanced cancer [PubMed: 25113111]. The therapeutic application of this epitope is restricted to patients who are HLA-A*2402 positive, making HLA typing a critical biomarker for patient selection. Its high tumor specificity helps minimize off-target effects, although common side effects include injection site reactions and low-grade fever.
