The URLC10 – HLA-A*24:02 complex consists of a specific peptide derived from the Up-regulated in lung cancer 10 (URLC10) protein, also known as Lymphocyte Antigen 6 Family Member K (LY6K), presented by the Human Leukocyte Antigen (HLA) allele A*24:02 (PubMed: 15150592). URLC10 is a cancer-testis antigen that is highly expressed in various malignancies, including non-small cell lung cancer and esophageal squamous cell carcinoma, while remaining largely silent in normal adult tissues except for the testis (UniProt: Q9BYG4). This restricted expression pattern makes the complex an attractive target for immunotherapy, as it allows for the selective destruction of cancer cells by the immune system. Therapeutic strategies targeting this complex include peptide-based vaccines and T-cell receptor (TCR) engineered T-cell therapies, which aim to induce a robust cytotoxic T-lymphocyte (CTL) response against tumors (ClinicalTrials.gov: NCT01213407). Because the HLA-A*24:02 allele is highly prevalent in East Asian populations, this target is particularly relevant for precision oncology in those regions.
