Urea transporter 2 (UT-A2) is a transmembrane protein encoded by the SLC14A2 gene, primarily expressed in the thin descending limb of the loop of Henle within the kidney [1][2]. It facilitates the passive transport of urea, which is a critical component of the intrarenal urea recycling process [2]. This recycling is essential for establishing and maintaining the high osmotic gradient in the inner medulla, which allows for efficient water reabsorption and the production of concentrated urine [3]. From a therapeutic perspective, UT-A2 is considered a promising target for "urearetic" diuretics [4]. Unlike traditional diuretics that target sodium transporters and often cause electrolyte imbalances, UT-A2 inhibitors promote water excretion by disrupting the medullary osmotic gradient [4][5]. Research into small-molecule inhibitors of UT-A2 is currently focused on treating fluid overload states associated with congestive heart failure, cirrhosis, and nephrotic syndrome [5].
