The bladder urothelium protective barrier refers to the specialized multilayered epithelial lining of the urinary bladder that safeguards underlying tissues from urine components, toxins, and microbes. This barrier is formed by several molecular and structural elements: the apical surface is covered by umbrella cells with impermeable membranes and uroplakin plaques, intercellular tight junctions restrict paracellular passage, and a glycocalyx rich in glycosaminoglycans provides anti-adherence and antibacterial properties. Compromise of this barrier leads to painful, inflammatory, or infectious bladder conditions. It is not a single molecular entity but a complex, dynamic interface with roles in protection, sensory signaling, and disease.
