Overview

The Vascular endothelial growth factor A (VEGF-A) gene promoter is a critical regulatory DNA sequence located upstream of the VEGFA coding region that controls the expression of the VEGF-A protein, a master regulator of angiogenesis (Semenza, 2003). This promoter region contains several essential regulatory elements, most notably the hypoxia-response element (HRE), which binds hypoxia-inducible factor 1 (HIF-1) to trigger vessel growth under low-oxygen conditions (PubMed: 12847480). Additionally, the promoter is characterized by G-rich sequences capable of forming G-quadruplex (G4) structures, which act as molecular switches to modulate transcriptional activity (Sun et al., 2005). In pathological states such as cancer, the VEGF-A promoter is often overactivated, facilitating tumor progression and metastasis by ensuring a robust blood supply (Nature Reviews Cancer, 2002). Conversely, insufficient activation of this promoter is linked to ischemic conditions and impaired wound healing in diabetic patients. Therapeutic strategies targeting this DNA region include engineered zinc finger transcription factors, such as SB-509, designed to activate expression for treating neuropathy, as well as small-molecule G-quadruplex stabilizers intended to repress transcription in oncology settings.
