Vertebrae development associated protein (Vertnin, VRTN) is a nuclear DNA-binding transcription factor conserved in mammals which regulates the development of thoracic vertebrae[7][2]. It acts during embryogenesis, where it is specifically expressed at the thoracic-somite stage and is indispensable for formation and segmentation of thoracic vertebrae and ribs. In animal models, VRTN modulates the segmentation clock through the Notch signaling pathway, coordinating somite formation and vertebral segmentation[1]. Variants in VRTN influence vertebral number in livestock, with significant agricultural impact. In mice, homozygous deficiency causes embryonic lethality while heterozygous animals may show abnormal spinal development and kyphosis[1]. The canonical molecular role of VRTN is as a transcription factor rather than a classical therapeutic target; no drugs are known to modulate its function and no validated clinical biomarkers or safety modalities exist for direct modulation[2][7].
