Vesicle-associated membrane protein 2 (VAMP2), also known as Synaptobrevin-2, is a critical v-SNARE protein primarily localized on the membranes of synaptic vesicles (UniProt P63027). It plays an essential role in neurotransmission by forming a stable SNARE complex with syntaxin-1 and SNAP-25, which facilitates the fusion of vesicles with the presynaptic membrane to release neurotransmitters (Wikipedia, 2024). Beyond its canonical role in the central nervous system, VAMP2 is involved in various secretory processes, including the translocation of GLUT4 in adipocytes and the recycling of specific G protein-coupled receptors (Rockefeller University Press, 2023). Mutations in the VAMP2 gene are associated with severe neurodevelopmental disorders characterized by intellectual disability, hypotonia, and epilepsy (PMC6451736). Clinically, VAMP2 is the primary molecular target for Botulinum neurotoxin type B (RimabotulinumtoxinB), which proteolytically cleaves the protein to inhibit acetylcholine release, providing therapeutic relief for conditions like sialorrhea and cervical dystonia (Patsnap Synapse, 2024). Additionally, VAMP2 levels in the cerebrospinal fluid are being investigated as potential biomarkers for synaptic degeneration in Alzheimer's disease and other neurodegenerative conditions (PMC8814781).
Proteolytic cleavage of the VAMP2 protein, specifically at the Gln76-Phe77 bond, which prevents the assembly of the SNARE complex and inhibits the fusion of synaptic vesicles with the presynaptic membrane, thereby blocking neurotransmitter release.
