Overview

Vestibular nuclei neurons are a specialized group of nerve cells located in the brainstem (medulla and pons) that serve as the primary processing center for balance and spatial orientation (StatPearls, 2023). They integrate sensory inputs from the vestibular apparatus of the inner ear with visual and somatosensory information to control the vestibulo-ocular reflex (VOR) and maintain postural equilibrium (NIH, 2022). In clinical practice, these neurons are the functional targets for drugs treating vertigo and motion sickness, as they express high levels of histamine H1, muscarinic, and GABA receptors (PubMed, 2021). Pathological activity or damage to these neurons results in debilitating symptoms such as vertigo, nausea, and ataxia (Wikipedia, 2024). While the term refers to a cellular population rather than a single molecule, it is a central focus in neuropharmacology for managing vestibular dysfunction (Journal of Vestibular Research, 2020).

Mechanism of action

Pharmacological agents modulate these neurons by acting as Histamine H1 receptor antagonists, Muscarinic acetylcholine receptor antagonists, or GABA-A receptor agonists to suppress overactive vestibular signaling (StatPearls, 2023; PubMed, 2021).
