Viral DNA polymerase is a key enzyme encoded by herpesviruses, essential for viral genome replication during the lytic phase of infection. It belongs to the B family of DNA polymerases and is a primary target for antiviral drugs. The enzyme typically functions as part of a two-subunit complex, responsible for high-fidelity synthesis of viral genomic DNA. It possesses DNA polymerase, 3′–5′ exonuclease proofreading, and ribonuclease H activities. All current anti-herpetic drugs targeting HSV or HCMV act on their respective viral DNA polymerases.
