Overview

The viral envelope membrane of SARS-CoV-2 is a lipid bilayer derived from the host cell, into which three key structural proteins are embedded: spike (S), envelope (E), and membrane (M) proteins. This envelope is essential for the virus’s structure, infectivity, assembly, and interaction with host cells. It consists mainly of phospholipids with very little cholesterol or sphingolipids. Procoagulant amino-phospholipids are present on the external side at higher levels than in activated platelets. The envelope membrane provides a protective barrier for the viral RNA genome and facilitates entry into host cells via fusion events mediated by S protein. E and M proteins regulate intracellular trafficking of S protein as well as its processing during virion maturation. E protein’s ion channel activity may modulate virus-host interactions and contribute to pathogenicity. The unique lipid composition contributes directly to its ability to promote blood coagulation. Both M and E proteins are targets for patient antibodies, making them potential candidates for vaccine or therapeutic development.
