Target intelligence / Profile preview
Voltage-gated potassium (Kv) channels are a large family of transmembrane proteins that are fundamental to the regulation of electrical activity in neurons (StatPearls: Physiology, Voltage-Gated Potassium Channels [1]). These channels open in response to membrane depolarization, allowing the selective outward flow of potassium ions, which is the primary mechanism for action potential repolarization and the maintenance of the resting membrane potential (UniProt: Potassium voltage-gated channel subfamily A member 1 [2]). By controlling the duration and frequency of neuronal firing, Kv channels serve as critical determinants of synaptic transmission and neuronal excitability (PubMed: Voltage-gated potassium channels as therapeutic targets [3]). Dysfunction of these channels, often due to genetic mutations in subunits like KCNQ2 or KCNA1, is associated with various neurological disorders, including epilepsy, episodic ataxia, and neuropathic pain (PubMed: Potassium channels in health and disease [5]). Pharmacological agents target these channels to either dampen hyperexcitability, as seen with the opener retigabine in epilepsy treatment, or to enhance conduction, as seen with the blocker dalfampridine in multiple sclerosis (PubChem: Retigabine [4]). However, therapeutic development is often challenged by the need for subunit selectivity to avoid adverse effects, most notably the risk of cardiac arrhythmias caused by off-target inhibition of the hERG channel (PubMed: The hERG potassium channel and drug-induced QT prolongation [6]).
Drugs targeting these channels function as either channel openers (activators) that enhance potassium efflux to hyperpolarize the membrane and reduce excitability, or as channel blockers (antagonists) that prolong the action potential to improve nerve conduction (PubMed: Voltage-gated potassium channels as therapeutic targets [3]).
