Whole inactivated SARS-CoV-2 virion antigen (including spike glycoprotein)

Molecular classification
Other, Viral structural proteins, Class I viral fusion protein
01

Overview

The whole inactivated SARS-CoV-2 virion antigen is a preparation of SARS-CoV-2 virus particles rendered non-infectious through chemical or physical methods. These particles retain the structural proteins found in the natural virus, most notably the spike glycoprotein (S protein), which mediates viral entry into host cells and is the primary target of neutralizing antibodies[1][2][3][4][6][7]. Other components include the nucleocapsid (N protein), membrane (M protein), and envelope (E protein). As an antigen source for vaccines, these virions present multiple epitopes, eliciting a broad immune response. Therapeutically, the terminology does not denote a classic drug target such as a receptor or an enzyme, but rather the antigenic stimulus used to train the immune system to recognize and combat SARS-CoV-2 infection. Note: The entry conflates multiple distinct viral proteins and should not be used as a canonical molecular target for drug discovery or pharmacological profiling. For mechanistic or drug interaction data, focus should be specifically on the spike glycoprotein, which is the principal target for neutralizing antibodies and entry inhibitors[1][3][4][5][6][7], or other individual SARS-CoV-2 proteins.

Mechanism of action

Used as a vaccine antigen: stimulates a broad immune response by presenting multiple viral proteins, mainly neutralizing antibodies against the spike glycoprotein, and T-cell responses to other proteins. Not a drug target for direct therapeutic inhibition.

03

Biological functions

Viral entry into host cells (primarily spike glycoprotein)Induction of host immune response (as antigens in vaccines or infection)
04

Disease associations

Infection (COVID-19; causative agent)
05

Safety considerations

Rare risk of vaccine-associated enhanced disease (VAED) observed with some whole virion vaccines in prior coronavirus studiesPossible allergic reaction to contaminants or additives in vaccine formulationsStandard vaccine safety concerns (local/injection site reactions, fever)

07

Biomarkers

Anti-spike protein antibody titers (as a marker of vaccine response or prior infection)Anti-nucleocapsid antibody titers (distinguishes infection from spike-only vaccine response)
