Overview

The Wnt family member 3A (WNT3A) messenger RNA 3'-untranslated region (3'-UTR) is a critical regulatory segment of the WNT3A transcript that controls the stability and translation of the encoded protein (UniProt P56704). WNT3A is a secreted glycoprotein that initiates the canonical Wnt/beta-catenin signaling pathway, which is essential for cell fate determination and tissue homeostasis (PubMed PMID: 21822264). In various malignancies, such as colorectal and hepatocellular carcinoma, WNT3A is frequently overexpressed due to the loss of post-transcriptional regulation at the 3'-UTR (PubMed PMID: 24954369). This region contains binding sites for microRNAs, such as miR-203 and miR-30, which act as endogenous suppressors of WNT3A expression (PubMed PMID: 25633788). Consequently, the WNT3A mRNA 3'-UTR is an emerging therapeutic target for RNA-based interventions, including microRNA mimics and antisense oligonucleotides (ASOs), designed to silence oncogenic Wnt signaling. By reducing WNT3A protein levels, these therapies aim to inhibit tumor growth and metastasis in Wnt-dependent cancers. However, therapeutic application faces challenges such as ensuring tissue-specific delivery and avoiding systemic toxicity associated with broad Wnt pathway inhibition (PubMed PMID: 28611104).
