Overview

X antigen family member 1B (XAGE-1b) is a prominent member of the cancer-testis antigen (CTA) family, characterized by its restricted expression in normal germ cells of the testis and its aberrant overexpression in various cancers, particularly lung adenocarcinoma (UniProt P56955). It is highly immunogenic, frequently triggering both humoral and cellular immune responses in patients with XAGE-1b-positive tumors (PubMed: 22496204). In the context of non-small cell lung cancer (NSCLC), XAGE-1b expression is associated with specific clinical outcomes and serves as a potential target for therapeutic vaccines and T-cell-based therapies (PubMed: 25193440). Its biological role in cancer involves promoting cell survival and proliferation, making it a functional target beyond its role as an immunological marker. Current clinical research focuses on utilizing XAGE-1b as a biomarker for patient stratification and as a target for peptide-based or mRNA-based immunotherapy (PubMed: 30104315).
