Zinc finger protein, FOG family member 1 (ZFPM1), also known as Friend of GATA 1 (FOG1), is a transcription regulator essential for the development of erythroid (red blood) and megakaryocytic (platelet-forming) cell lineages[1][4][5][11]. ZFPM1 functions primarily as a transcriptional cofactors by forming heterodimers with GATA family transcription factors (including GATA1, GATA2, and GATA3), modulating their activity to either activate or repress specific gene expression programs depending on cell and promoter context[1][4][5][11]. ZFPM1 is thus pivotal in regulating genes required for differentiation and function of blood cells, with mutations or deficiencies linked to blood disorders such as congenital dyserythropoietic anemia and thrombocytopenia[1][4][11]. Structurally, ZFPM1 contains multiple zinc finger domains mediating protein-DNA and protein-protein interactions[3]. It is mainly localized to the cell nucleus and exhibits broad tissue expression, especially in hematopoietic tissues[6]. No drugs are currently approved targeting ZFPM1 directly, but its central regulatory role in hematopoiesis marks it as a potential therapeutic target[4].
