Overview

Zinc finger protein ZIC 5 (ZIC5) is a member of the ZIC family of C2H2-type zinc finger transcription factors, which are essential for early embryonic development, particularly in neural crest specification and central nervous system patterning (UniProt: Q96FZ8). While its expression is typically restricted in healthy adult tissues, ZIC5 mRNA is frequently overexpressed in various malignancies, including melanoma, colorectal cancer, and non-small cell lung cancer, where it acts as a potent oncogene (PubMed: 23418324). It promotes tumor progression by driving key signaling pathways such as Wnt/beta-catenin and Sonic Hedgehog, which facilitate cell proliferation, migration, and resistance to apoptosis (PubMed: 30106434). Because transcription factors like ZIC5 are traditionally difficult to target with small molecules, therapeutic strategies have focused on silencing the ZIC5 mRNA transcript using RNA interference (RNAi) or antisense oligonucleotides (ASOs). Preclinical studies have demonstrated that reducing ZIC5 mRNA levels can effectively inhibit tumor growth and metastatic potential in vivo (PubMed: 28656244). Consequently, ZIC5 is considered a promising target for nucleic acid-based precision medicine, although clinical translation requires overcoming challenges related to targeted delivery and the mitigation of off-target effects.
