Overview
Alethia’s lead antibody sotevtamab (formerly AB-16B5) targets EMT by inhibiting secreted clusterin. In preclinical models, it reduced tumor invasion and improved chemotherapy response. Phase I trials showed safety/tolerability with stable disease in several patients up to one year. A completed open-label Phase II trial in NSCLC demonstrated efficacy signals; a new proof-of-concept Phase II trial in colorectal cancer with liver metastases has been initiated. Recent data from the EGIA-002 trial were presented at ASCO 2024.
