Overview
Olema’s lead candidate palazestrant (OP-1250), a complete estrogen receptor antagonist and selective ER degrader, has shown promising efficacy and tolerability in early clinical trials for metastatic breast cancer. The pivotal Phase 3 OPERA‑01 trial as monotherapy for second/third-line ER+/HER2– metastatic breast cancer is ongoing with top-line data expected in 2026. A second pivotal trial, OPERA‑02, will test palazestrant with ribociclib as first-line therapy. Early-phase combination data show enhanced anti-tumor activity. The company’s second asset, OP‑3136—a KAT6 inhibitor—entered Phase 1 trials at the end of 2024 targeting multiple solid tumors including breast cancer
