Pellficure Pharmaceuticals developed PCUR-101, an oral synthetic plumbagin derivative targeting STAT3 for metastatic castration-resistant prostate cancer. Two Phase I studies were conducted combining PCUR-101 with androgen deprivation therapy. The drug was generally well tolerated at tested doses; most adverse events were mild gastrointestinal symptoms. No dose-limiting toxicities or objective responses were seen, but some patients achieved stable disease and PSA reduction. Trials were terminated to allow reformulation for higher dosing and to redevelop pharmacokinetic assays; there are no reports of resumed development as of March 2025[4][5].
