Quanta Therapeutics is conducting Phase 1 clinical trials on multiple oral KRAS inhibitors targeting KRAS G12D and KRAS G12V mutations in difficult-to-treat cancers. Their leading candidate, QTX3034 (G12D+ multi-KRAS inhibitor), is in dose expansion cohorts in KRAS G12D-mutant pancreatic, colorectal, and endometrial cancers. QTX3544 (G12V+ multi-KRAS inhibitor) is in Phase 1 for KRAS G12V-mutant solid tumors, including planned combination therapy with cetuximab for enhanced efficacy. Early clinical data show favorable safety and encouraging efficacy. The platform allows real-time detection of protein conformational changes, supporting the development of RAS-targeted small molecules that may overcome resistance seen with earlier-generation KRAS inhibitors
