TeGenero Immuno Therapeutics

TeGenero Immuno Therapeutics was a German biotechnology company specializing in the development of novel immunotherapies based on superagonistic monoclonal antibodies. The company's lead candidate, TGN1412, was designed to directly activate T-cells by targeting the CD28 receptor, intended for the treatment of autoimmune diseases and hematological malignancies. TeGenero became globally recognized following a disastrous Phase 1 clinical trial in March 2006 at Northwick Park Hospital, where all six healthy volunteers suffered near-fatal cytokine release syndrome. The clinical failure and subsequent loss of funding led the company to file for insolvency in July 2006. The incident remains a landmark case in drug safety, leading to fundamental changes in how first-in-human trials for high-risk biologics are regulated.
