Thermally Targeted Therapeutics is a preclinical-stage biotechnology company developing a proprietary drug delivery platform based on genetically engineered, thermally responsive elastin-like polypeptides (ELPs). The company's technology leverages the unique properties of ELPs, which remain soluble at normal physiological temperatures but undergo a phase transition to form aggregates in response to mild, externally applied hyperthermia (40–41°C). By conjugating therapeutic peptides or small-molecule chemotherapeutics to these ELPs, along with cell-penetrating peptides (CPPs), the company achieves highly localized drug accumulation and enhanced cellular uptake at the tumor site. This triple-targeting approach (combining passive EPR accumulation, active thermal targeting, and CPP-mediated internalization) is designed to maximize therapeutic efficacy while minimizing systemic toxicities in indications such as glioblastoma, prostate cancer, breast cancer, and pancreatic cancer.
