Tilikum Therapeutics has developed a novel class of PARP-locking small molecule inhibitors that show strong cytotoxicity against cancer cells at low doses. Unlike conventional PARP inhibitors that require pre-existing DNA repair defects in tumors, Tilikum’s compounds are active even in tumors without such defects or those with acquired resistance. Their lead compound ("TLK001", also referred to as "Pip6") has demonstrated promising preclinical efficacy and will be profiled in upcoming Phase 1A/B trials targeting high-replicative stress tumor populations.
