vTv Therapeutics’ lead asset cadisegliatin (also known as TTP399) is a novel oral small molecule liver-selective glucokinase activator being developed as an adjunct to insulin for type 1 diabetes. The drug has shown a ~40% reduction in hypoglycemic episodes versus placebo in phase II and demonstrated no increased risk of ketoacidosis. Cadisegliatin received FDA Breakthrough Therapy designation and entered the pivotal CATT1 phase III trial; after a temporary clinical hold due to an analytical signal concern that was resolved by March–May of 2025, the trial resumed with topline data expected in H2 of 2026. The company’s pipeline also includes earlier-stage or partnered programs targeting metabolic diseases such as type II diabetes and cystic fibrosis-related diabetes.
