α-SC9 is an experimental recombinant protein vaccine candidate designed for the treatment of synucleinopathies, including Parkinson's disease (PD), multiple system atrophy (MSA), and dementia with Lewy bodies (DLB). Developed using a rational design approach, the vaccine utilizes the prion domain of the fungal HET-s protein as a rigid structural scaffold. Specific epitopes from the surface of alpha-synuclein amyloid fibrils are grafted onto this scaffold to mimic the parallel in-register beta-sheet structure characteristic of pathological aggregates. This design allows the immune system to generate high-affinity IgG antibodies that selectively recognize and target misfolded alpha-synuclein fibrils while avoiding cross-reactivity with native, monomeric alpha-synuclein, thereby reducing the risk of harmful autoimmune responses.
