α3nAChR siRNA is a small interfering RNA (siRNA) designed to specifically silence the expression of the alpha 3 subunit of the nicotinic acetylcholine receptor (α3nAChR), which is encoded by the CHRNA3 gene. In the context of Alzheimer's disease research, this tool is utilized to investigate the regulatory role of α3nAChR in the amyloidogenic pathway. Experimental evidence in human neuroblastoma SH-SY5Y cells indicates that silencing α3nAChR leads to the downregulation of key components involved in amyloid precursor protein (APP) processing, such as ADAM10 (α-secretase), presenilin 1 (PS1), and nicastrin (NCT), as well as effectors in the Notch signaling pathway (Notch1 and Hes1). These findings suggest that α3nAChR may promote the non-amyloidogenic processing of APP and exert a protective effect against β-amyloid (Aβ) production.
