Drug intelligence / Profile preview
An α4β7-blocking monoclonal antibody is a class of therapeutic biologic agents designed to selectively target and inhibit the α4β7 integrin heterodimer, a cell-surface glycoprotein expressed on lymphocytes. By blocking the interaction between α4β7 integrin and its primary ligand, mucosal vascular addressin cell adhesion molecule 1 (MAdCAM-1), which is highly expressed on the vascular endothelium of the gastrointestinal tract, these antibodies prevent the homing and migration of inflammatory lymphocytes into gut tissues. This gut-selective mechanism of action reduces local inflammation without causing systemic immunosuppression. The most prominent approved drug in this class is vedolizumab (Entyvio), which is widely used for the treatment of moderate-to-severe inflammatory bowel diseases, including ulcerative colitis and Crohn's disease. Other candidates in this class include abrilumab (AMG 181) and SPY001.
