The α9α10 nAChR program, developed by Rapport Therapeutics in collaboration with Janssen Pharmaceuticals, focuses on the discovery and development of small molecule agonists targeting the α9α10 nicotinic acetylcholine receptor (nAChR). This receptor complex is highly enriched in the cochlear hair cells of the inner ear and plays a pivotal role in the olivocochlear pathway, which provides protective feedback against noise-induced damage. Rapport's proprietary Receptor Associated Proteins (RAP) platform technology enabled the functional expression and identification of these agonists, which were previously difficult to target using standard recombinant cell lines. The program aims to produce oral therapeutics with high selectivity and low nanomolar potency to treat conditions such as age-related hearing loss, acoustic trauma, tinnitus, and potentially vestibular disorders.
