Overview

αBCMA-CAR.CD19BiTE is an experimental chimeric antigen receptor (CAR) T-cell therapy engineered to target B-cell maturation antigen (BCMA) while simultaneously secreting a bispecific T-cell engager (BiTE) directed against CD19. Developed as a research tool and control construct by investigators at the Huntsman Cancer Institute and Dana-Farber Cancer Institute, this dual-targeting approach aims to address antigen escape and enhance the efficacy of CAR T-cell therapy in hematologic malignancies like multiple myeloma. While BCMA is a primary target in myeloma, the secretion of a CD19 BiTE serves to recruit and activate bystander T cells against CD19-positive cells, although in the context of the provided research, it primarily serves as a negative control for a CD38-secreting variant.
