Drug intelligence / Profile preview
αC-targeting Adhirons are small engineered protein binders (~12 kDa) based on a plant cystatin scaffold (previously known as Affimers) that selectively target the intrinsically disordered αC-region of fibrinogen with high affinity. Developed by researchers at the University of Leeds, these binders (including specific candidates αC-5, αC-8, and αC-45) modulate fibrin clot structure and stability. They exhibit dual effects on haemostasis and thrombosis: during clot formation, they delay clotting and decrease clot firmness (suggesting an antithrombotic effect), whereas once the clot is formed, they prolong clot stability and decelerate fibrinolysis. They are being investigated as potential therapeutic tools for modulating clot properties and reducing abnormal clot formation during thrombosis or bleeding complications.
