αCD123 CAR-T is a chimeric antigen receptor (CAR) T-cell therapy designed to target CD123 (Interleukin-3 receptor subunit alpha), an antigen frequently overexpressed on the surface of leukemic blasts and leukemic stem cells in acute myeloid leukemia (AML). The specific construct described in preclinical research by the Hospital Clinic de Barcelona utilizes a second-generation CAR architecture comprising a single-chain variable fragment (scFv) targeting CD123, a CD28 costimulatory domain, and a CD3ζ signaling domain. This therapeutic approach aims to provide a potent cytotoxic response against AML cells, potentially serving as a bridge to allogeneic hematopoietic stem cell transplantation for patients with relapsed or refractory disease.
